Genes affected
WWOX (HGNC:12799): (WW domain containing oxidoreductase) This gene encodes a member of the short-chain dehydrogenases/reductases (SDR) protein family. This gene spans the FRA16D common chromosomal fragile site and appears to function as a tumor suppressor gene. Expression of the encoded protein is able to induce apoptosis, while defects in this gene are associated with multiple types of cancer. Disruption of this gene is also associated with autosomal recessive spinocerebellar ataxia 12. Disruption of a similar gene in mouse results in impaired steroidogenesis, additionally suggesting a metabolic function for the protein. Alternative splicing results in multiple transcript variants. This nucleotide change is present in gnomAD database with a low allele frequency of 0.0011% [PM2]. Insilico prediction [MutationTaster] predicts a deleterious nature of this variant [PP3]. A clinvar entry [Variation id: 241105] of this variant is already present with a "Pathogenic" interpretation [PP5]. Pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpDec 20, 2023This sequence change creates a premature translational stop signal (p.Arg264*) in the WWOX gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in WWOX are known to be pathogenic (PMID: 24456803, 25411445). This variant is present in population databases (rs756762196, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with WWOX-related conditions. ClinVar contains an entry for this variant (Variation ID: 241105). For these reasons, this variant has been classified as Pathogenic. -

Pathogenic, no assertion criteria providedclinical testingPreventionGenetics, part of Exact SciencesApr 29, 2024The WWOX c.790C>T variant is predicted to result in premature protein termination (p.Arg264*). This variant has been reported in the homozygous state in individuals with WWOX-related epileptic encephalopathy (Supplemental Table 1, Piard et al 2019. PubMed ID: 30356099; Iacomino M et al 2020. PubMed ID: 32581702) and the heterozygous state in individuals with epilepsy (Supplemental Table 2, Truty R et al 2019. PubMed ID: 31440721). This variant is reported in 0.0065% of alleles in individuals of South Asian descent in gnomAD. Nonsense variants in WWOX are expected to be pathogenic. This variant is interpreted as pathogenic. -
not provided Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingGeneDxAug 10, 2015The R264X variant in the WWOX gene has not been reported previously as a germline pathogenic variant nor as a benign polymorphism, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The R264X variant was not observed in approximately 6,100 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret R264X as a pathogenic variant. -
