Truncations downstream of this position have been classified as likely pathogenic by our laboratory, and others are found in association with Brittle Cornea Syndrome in HGMD. The variant allele was found at a frequency of 0.00019 in 125018 control chromosomes. This frequency is not significantly higher than expected for a pathogenic variant in ZNF469 causing Brittle Cornea Syndrome 1 (0.00019 vs 0.0011). To our knowledge, no occurrence of c.19C>T in individuals affected with Brittle Cornea Syndrome 1 and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
