The protein encoded by this gene is a component of both U2- and U12-dependent spliceosomes, and found to be essential for the catalytic step II in pre-mRNA splicing process. It contains several WD repeats, which function in protein-protein interactions. This protein has a sequence similarity to yeast Prp8 protein. This gene is a candidate gene for autosomal dominant retinitis pigmentosa. [provided by RefSeq, Jul 2008]

This sequence change replaces tyrosine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 2334 of the PRPF8 protein (p.Tyr2334Asn). This missense change has been observed in individuals with clinical features of autosomal dominant retinitis pigmentosa (PMID: 20232351, 21378395; Invitae). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 865924). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). Experimental studies have shown that this missense change affects PRPF8 function (PMID: 28515276). For these reasons, this variant has been classified as Pathogenic. -
