The protein encoded by this gene is an integral membrane protein and inward-rectifier type potassium channel. The encoded protein, which tends to allow potassium to flow into rather than out of a cell, can form heterodimers with two other inward-rectifier type potassium channels. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0035% of alleles in individuals of European (Non-Finnish) descent in gnomAD (http://gnomad.broadinstitute.org/variant/17-68128695-T-G). Other nonsense variants in KCNJ16 have been reported in patients with hypokalemic metabolic acidosis and sensorineural deafness phenotypes (see, for example, Webb et al. 2021. PubMed ID: 33840812; Schlingmann et al. 2021. PubMed ID: 33811157 ). Taken together, we interpret this variant as likely pathogenic. - |
