The protein encoded by this gene is an integral membrane protein and inward-rectifier type potassium channel. The encoded protein, which tends to allow potassium to flow into rather than out of a cell, can form heterodimers with two other inward-rectifier type potassium channels. Other nonsense variants in KCNJ16 have been reported in patients with hypokalemic metabolic acidosis and sensorineural deafness phenotypes (see, for example, Webb et al. 2021. PubMed ID: 33840812; Schlingmann et al. 2021. PubMed ID: 33811157 ). Taken together, we interpret this variant as likely pathogenic. -
