This variant is predicted to cause loss of normal protein function through protein truncation. The c.1555delA variant is observed in 9/10,150 (0.088%) alleles from individuals of Ashkenazi Jewish background, in the ExAC dataset (Lek et al., 2016). The variant allele was found at a frequency of 0.00016 in 251448 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in CCDC8 causing Three M Syndrome 3 (0.00016 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1555delA in individuals affected with Three M Syndrome 3 and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
