This variant is present in population databases (rs745914961, gnomAD 0.09%). This variant has not been reported in the literature in individuals affected with CCDC8-related conditions. ClinVar contains an entry for this variant (Variation ID: 452737). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The c.1555delA variant causes a frameshift starting with codon Arginine 519, changes this amino acid to a Glycine residue, and creates a premature Stop codon at position 3 of the new reading frame, denoted p.Arg519GlyfsX3. This variant is predicted to cause loss of normal protein function through protein truncation. The c.1555delA variant is observed in 9/10,150 (0.088%) alleles from individuals of Ashkenazi Jewish background, in the ExAC dataset (Lek et al., 2016). We interpret c.1555delA as a variant of uncertain significance. -

The variant allele was found at a frequency of 0.00016 in 251448 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in CCDC8 causing Three M Syndrome 3 (0.00016 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1555delA in individuals affected with Three M Syndrome 3 and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
