The V674L variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The V674L variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is not conserved, and in silico analysis predicts this variant likely does not alter the protein structure/function. We interpret V674L as a variant of uncertain significance. -

This variant is present in population databases (rs761108884, ExAC 0.01%). This sequence change replaces valine with leucine at codon 674 of the SLC25A12 protein (p.Val674Leu). The valine residue is weakly conserved and there is a small physicochemical difference between valine and leucine. -
