The Asp. at codon 3 is replaced by Asn , This amino acid position is not conserved (PhyloP= -0.220 ). This variant not present in our local database nor was it identified in the Genome Aggregation Database The computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) do not suggest a high likelihood the variant Asn impacts the protein; however, this information is not predictive enough to rule out pathogenicity. ClinVar has an entry for this variant (438899) as uncertain significance submitted by four different diagnostic labs . Since supporting evidence is limited at this time, this variant is classified as of uncertain significance. -
