The variant was absent in 243692 control chromosomes (gnomAD). c.331C>T has been reported in the literature in individuals affected with Primary Hyperoxaluria Type 1 (examples: Du_2018, and Birtel_2019). At least one publication reports experimental evidence that this variant leads to absence of protein and enzyme activity (Du_2018). The following publications have been ascertained in the context of this evaluation (PMID: 31078535, 30341509). Two submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
