The encoded protein assembles and transfers 4Fe-4S clusters to target apoproteins including succinate dehydrogenase and lipoic acid synthase. Mutations in this gene are a cause of multiple mitochondrial dysfunctions syndrome-1, and pseudogenes of this gene are located on the short arms of chromosomes 1 and 3. One in silico tool, TraP, predicts it is possibly pathogenic. The variant was absent in 234624 control chromosomes (gmomAD). To our knowledge, no occurrence of c.721-1G>A in individuals affected with Multiple Mitochondrial Dysfunctions Syndrome 1 and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance. - |
