Inborn genetic diseases Uncertain:1
The c.2438G>A (p.R813H) alteration is located in exon 19 (coding exon 19) of the ADAM17 gene. This alteration results from a G to A substitution at nucleotide position 2438, causing the arginine (R) at amino acid position 813 to be replaced by a histidine (H). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Inflammatory skin and bowel disease, neonatal, 1 Uncertain:1
This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 813 of the ADAM17 protein (p.Arg813His). This variant is present in population databases (rs760757602, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with ADAM17-related conditions. ClinVar contains an entry for this variant (Variation ID: 961964). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Not Available"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
