Genes affected
PDYN (HGNC:8820): (prodynorphin) The protein encoded by this gene is a preproprotein that is proteolytically processed to form the secreted opioid peptides beta-neoendorphin, dynorphin, leu-enkephalin, rimorphin, and leumorphin. These peptides are ligands for the kappa-type of opioid receptor. Dynorphin is involved in modulating responses to several psychoactive substances, including cocaine. Multiple alternatively spliced transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2010]
PDYN-AS1 (HGNC:53462): (PDYN antisense RNA 1)

Likely pathogenic, criteria provided, single submitterclinical testingInvitaeJan 08, 2024This sequence change creates a premature translational stop signal (p.Trp220Glyfs*33) in the PDYN gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 35 amino acid(s) of the PDYN protein. This variant is present in population databases (rs748307861, gnomAD 0.01%). This premature translational stop signal has been observed in individuals with PDYN-related conditions (PMID: 23471613, 27528516). ClinVar contains an entry for this variant (Variation ID: 211896). Studies have shown that this premature translational stop signal alters PDYN gene expression (PMID: 23471613). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. -
