Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The molecular mechanism of disease for RBX1 is unknown at this time. The variant was absent in 251378 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.228+1G>T has been reported in an individual in internal testing. This report does not provide unequivocal conclusions about association of the variant with RBX1-Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
