Genes affected
CD96 (HGNC:16892): (CD96 molecule) The protein encoded by this gene belongs to the immunoglobulin superfamily. It is a type I membrane protein. The protein may play a role in the adhesive interactions of activated T and NK cells during the late phase of the immune response. It may also function in antigen presentation. Alternative splicing generates multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Jan 2016]

Uncertain significance, criteria provided, single submitterclinical testingInvitaeMay 17, 2021In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with CD96-related conditions. This variant is present in population databases (rs199773662, ExAC 0.02%). This sequence change replaces leucine with proline at codon 120 of the CD96 protein (p.Leu120Pro). The leucine residue is weakly conserved and there is a moderate physicochemical difference between leucine and proline. -
