However, in at least one of these families, a pathogenic CASR allele was also identified which suggests that this c.740C>T variant was not the primary cause of disease (PMID: 22192860, 24735972). ClinVar contains an entry for this variant (Variation ID: 64438). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
