APPL1 (HGNC:24035): (adaptor protein, phosphotyrosine interacting with PH domain and leucine zipper 1) The protein encoded by this gene has been shown to be involved in the regulation of cell proliferation, and in the crosstalk between the adiponectin signalling and insulin signalling pathways. The encoded protein binds many other proteins, including RAB5A, DCC, AKT2, PIK3CA, adiponectin receptors, and proteins of the NuRD/MeCP1 complex. The variant allele was found at a frequency of 0.00061 in 251212 control chromosomes, predominantly at a frequency of 0.0011 within the Non-Finnish European subpopulation in the gnomAD database, including 1 homozygote, strongly suggesting that the variant is a benign polymorphism found primarily in populations of Non-Finnish European origin. c.2018C>G has been reported in the literature in individuals affected with a clinical phenotype of neuronal ceroid lipofuscinosis (NCL), a genetically heterogeneous subtype of LSD (Di Fruscio_2015). The report does not provide unequivocal conclusions about association of the variant with Maturity-Onset Diabetes Of The Young Type 14.
