HINT1 (HGNC:4912): (histidine triad nucleotide binding protein 1) This gene encodes a protein that hydrolyzes purine nucleotide phosphoramidates substrates, including AMP-morpholidate, AMP-N-alanine methyl ester, AMP-alpha-acetyl lysine methyl ester, and AMP-NH2. The encoded protein interacts with these substrates via a histidine triad motif. This gene is considered a tumor suppressor gene. In addition, mutations in this gene can cause autosomal recessive neuromyotonia and axonal neuropathy. There are several related pseudogenes on chromosome 7. This variant disrupts the C-terminus of the HINT1 protein. Other variant(s) that disrupt this region (p.Gln106*, p.Trp123*) have been determined to be pathogenic (PMID: 25342199, 22961002, 29787766). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. This variant has not been reported in the literature in individuals with HINT1-related conditions. This variant is present in population databases (rs145306016, ExAC 0.006%). This sequence change results in a premature translational stop signal in the HINT1 gene (p.Arg95*). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 32 amino acids of the HINT1 protein. - |
