Genes affected
SERAC1 (HGNC:21061): (serine active site containing 1) The protein encoded by this gene is a phosphatidylglycerol remodeling protein found at the interface of mitochondria and endoplasmic reticula, where it mediates phospholipid exchange. The encoded protein plays a major role in mitochondrial function and intracellular cholesterol trafficking. Defects in this gene are a cause of 3-methylglutaconic aciduria with deafness, encephalopathy, and Leigh-like syndrome (MEGDEL). Two transcript variants, one protein-coding and the other non-protein coding, have been found for this gene. [provided by RefSeq, Aug 2012]

3-methylglutaconic aciduria with deafness, encephalopathy, and Leigh-like syndrome Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingInvitaeJun 09, 2022In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with SERAC1-related conditions. This variant is present in population databases (rs745780427, gnomAD 0.01%). This sequence change affects codon 613 of the SERAC1 mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the SERAC1 protein. -
