Variant summary: TNXB c.8536G>C (p.Gly2846Arg) results in a non-conservative amino acid change located in the Fibronectin type 3 domain (IPR003961) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00019 in 245848 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in TNXB causing Ehlers-Danlos syndrome due to tenascin-X deficiency (0.00019 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.8536G>C in individuals affected with Ehlers-Danlos syndrome due to tenascin-X deficiency and no experimental evidence demonstrating its impact on protein function have been reported.
