Genes affected
KCNH2 (HGNC:6251): (potassium voltage-gated channel subfamily H member 2) This gene encodes a component of a voltage-activated potassium channel found in cardiac muscle, nerve cells, and microglia. Four copies of this protein interact with one copy of the KCNE2 protein to form a functional potassium channel. Mutations in this gene can cause long QT syndrome type 2 (LQT2). Transcript variants encoding distinct isoforms have been identified. [provided by RefSeq, May 2022]

Pathogenic, criteria provided, single submitter | clinical testing | GeneDx | Mar 20, 2013 | Although the c.3104_3107dupGGGG variant in the KCNH2 gene has not been reported to our knowledge, this variant causes a shift in reading frame starting at codon Aspartic acid 1037, changing it to a Glycine, and creating a premature stop codon at position 83 of the new reading frame, denoted p.Asp1037GlyfsX83. This variant is expected to result in either an abnormal, truncated protein product or loss of protein from this allele through nonsense-mediated mRNA decay. Other frameshift variants in the KCNH2 gene have been reported in association with LQTS. In summary, c.3104_3107dupGGGG in the KCNH2 gene is interpreted as a pathogenic variant. - |
