Variant summary: KCNH2 c.551_568dup18 (p.Gly184_Gly189dup) results in an in-frame duplication that is predicted to duplicate six amino acids into the encoded protein. The variant was absent in 63692 control chromosomes (gnomAD). c.551_568dup18 has been reported in the literature in an individual referred for long QT syndrome genetic testing (Kapplinger_2009). This report does not provide unequivocal conclusions about association of the variant with Arrhythmia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
