This variant has been observed in individuals with breast cancer and stomach cancer (Park 2012, Lu 2015). Functional assays suggest this variant is able to restore RAD512C foci formation and restore homologous recombination efficiency similar to wild type (Hilbers 2016). XRCC2 Leu61Ile was not observed at a significant frequency in large population cohorts (Lek 2016). Since Leucine and Isoleucine share similar properties, this is considered a conservative amino acid substitution. XRCC2 Leu61Ile occurs at a position that is conserved across species and is not located in a known functional domain. In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available evidence, it is unclear whether XRCC2 Leu61Ile is pathogenic or benign. - |
