Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 22346 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. Loss of function variants in RAC1 have not been associated with Intellectual Disability, Autosomal Dominant 48 in HGMD. To our knowledge, no occurrence of c.35+2T>G in individuals affected with Intellectual Disability, Autosomal Dominant 48 and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014.
