This changes the amino acid from a tyrosine (Y) to a stop codon at amino acid position 765. This alteration is expected to result in premature protein truncation or nonsense-mediated mRNA decay. However, loss of function of AGO2 has not been clearly established as a mechanism of disease. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. - |
