The c.10A>T (p.N4Y) alteration is located in exon 1 (coding exon 1) of the CLN5 gene. This alteration results from a A to T substitution at nucleotide position 10, causing the asparagine (N) at amino acid position 4 to be replaced by a tyrosine (Y). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces asparagine with tyrosine at codon 4 of the CLN5 protein (p.Asn4Tyr). The asparagine residue is weakly conserved and there is a large physicochemical difference between asparagine and tyrosine. This variant has not been reported in the literature in individuals affected with CLN5-related conditions.
