The variant of interest has not been observed in controls (ExAC, 1000 Gs, or ESP), nor has it been, to our knowledge, reported in affected individuals via publications. The p.I975N variant (also known as c.2924T>A), located in coding exon 10 of the BRCA2 gene, results from a T to A substitution at nucleotide position 2924. The isoleucine at codon 975 is replaced by asparagine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
