In summary, this variant is a rare missense change with uncertain impact on protein function. There is no indication that it causes disease, but the available evidence is currently insufficient to prove that conclusively. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is present in population databases (rs770984577, ExAC 0.002%) but has not been reported in the literature in individuals with a FANCA-related disease. This sequence change replaces arginine with threonine at codon 32 of the FANCA protein (p.Arg32Thr). The arginine residue is weakly conserved and there is a moderate physicochemical difference between arginine and threonine. -

The p.R32T variant (also known as c.95G>C), located in coding exon 2 of the FANCA gene, results from a G to C substitution at nucleotide position 95. The arginine at codon 32 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
