Variant summary: FBN1 c.8480A>C (p.Tyr2827Ser) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251398 control chromosomes, however the available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.8480A>C in individuals affected with Marfan Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. Three ClinVar submitters provided clinical-significance assessments for this variant after 2014, all classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
The FBN1 c.8480A>C; p.Tyr2827Ser variant (rs794728287), to our knowledge, is not reported in the medical literature but is reported as uncertain significance in ClinVar (Variation ID: 200135). This variant is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The tyrosine at codon 2827 is moderately conserved, and computational analyses (SIFT, PolyPhen-2) predict that this variant may be deleterious. The p.Y2827S variant (also known as c.8480A>C), located in coding exon 65 of the FBN1 gene, results from an A to C substitution at nucleotide position 8480. The tyrosine at codon 2827 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

ClinVar contains an entry for this variant (Variation ID: 200135). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt FBN1 protein function with a positive predictive value of 95%.
