Hereditary nonpolyposis colorectal neoplasms Uncertain:1
In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with MSH6-related conditions. This variant is present in population databases (rs766557450, ExAC no frequency). This sequence change replaces lysine with threonine at codon 1233 of the MSH6 protein (p.Lys1233Thr). The lysine residue is moderately conserved and there is a moderate physicochemical difference between lysine and threonine. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.K1233T variant (also known as c.3698A>C), located in coding exon 8 of the MSH6 gene, results from an A to C substitution at nucleotide position 3698. The lysine at codon 1233 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
