Gastrointestinal stromal tumor Uncertain:1
This sequence change creates a premature translational stop signal (p.Gln947*) in the KIT gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 30 amino acid(s) of the KIT protein. This variant is present in population databases (no rsID available, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with KIT-related conditions. ClinVar contains an entry for this variant (Variation ID: 409760). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
Hereditary cancer-predisposing syndrome Uncertain:1
The p.Q947* variant (also known as c.2839C>T), located in coding exon 21 of the KIT gene, results from a C to T substitution at nucleotide position 2839. This changes the amino acid from a glutamine to a stop codon within coding exon 21. This alteration is expected to result in protein truncation or nonsense-mediated mRNA decay. However, loss of function of KIT has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
Mastocytosis;C0080024:Piebaldism Other:1
Variant classified as Uncertain significance and reported on 08-22-2022 by Invitae. GenomeConnect-InvitaePIN assertions are reported exactly as they appear on the patient-provided report from the testing laboratory. Registry team members make no attempt to reinterpret the clinical significance of the variant. Phenotypic details are available under supporting information.
