The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. The p.A1603T variant (also known as c.4807G>A), located in coding exon 32 of the MYH7 gene, results from a G to A substitution at nucleotide position 4807. The alanine at codon 1603 is replaced by threonine, an amino acid with similar properties. This alteration was reported in a cohort of individuals with hypertrophic cardiomyopathy; however, limited clinical information was provided (Millat G et al. Clin. Chim. Acta, 2010 Dec;411:1983-91). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
