The encoded protein also stabilizes constitutive heterochromatin to maintain the overall chromatin structure. The active, hypophosphorylated form of the protein binds transcription factor E2F1. The p.E322* pathogenic mutation (also known as c.964G>T), located in coding exon 10 of the RB1 gene, results from a G to T substitution at nucleotide position 964. This changes the amino acid from a glutamic acid to a stop codon within coding exon 10. This mutation was detected in two relatives diagnosed with retinoblastoma (Taylor M et al. Hum. Mutat. 2007 Mar;28(3):284-93). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
