Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 9585602, 19410294, 20729109). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Disruption of the initiator codon has been observed in individual(s) with chronic granulomatous disease (PMID: 29560547, 11162142, 22929960). This variant is not present in population databases (ExAC no frequency). This sequence change affects the initiator methionine of the CYBB mRNA. The next in-frame methionine is located at codon 65. -
