The p.I509T variant (also known as c.1526T>C), located in coding exon 6 of the BARD1 gene, results from a T to C substitution at nucleotide position 1526. The isoleucine at codon 509 is replaced by threonine, an amino acid with similar properties. This variant was reported in 1/196 familial breast cancer cases and in 0/245 controls. Four females in this family were diagnosed with breast cancer and the p.I509T variant co-segregated with the breast cancer (De Brakeleer S, et al. Hum. Mutat. 2010 Mar; 31(3):E1175-85). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
