This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with isoleucine at codon 602 of the BARD1 protein (p.Ser602Ile). The serine residue is moderately conserved and there is a large physicochemical difference between serine and isoleucine. -

The p.S602I variant (also known as c.1805G>T), located in coding exon 8 of the BARD1 gene, results from a G to T substitution at nucleotide position 1805. The serine at codon 602 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
