KCNJ2 (HGNC:6263): (potassium inwardly rectifying channel subfamily J member 2) Potassium channels are present in most mammalian cells, where they participate in a wide range of physiologic responses. The protein encoded by this gene is an integral membrane protein and inward-rectifier type potassium channel. The encoded protein, which has a greater tendency to allow potassium to flow into a cell rather than out of a cell, probably participates in establishing action potential waveform and excitability of neuronal and muscle tissues. Mutations in this gene have been associated with Andersen syndrome, which is characterized by periodic paralysis, cardiac arrhythmias, and dysmorphic features. [provided by RefSeq, Jul 2008]

The p.Asn410Ser variant (rs141069645) has been previously reported once in a cohort of arrhythmogenic right ventricular cardiomyopathy (AVRC) patients (Sabater-Molina 2013); however, specific genetic and clinical information about the patient in question was not reported. This variant is listed in the Genome Aggregation Database (gnomAD) browser with an overall frequency of 0.034% (identified in 94 out of 276,876 chromosomes) and is listed the ClinVar database as likely benign or as a variant of uncertain significance (Variation ID: 190804). The asparagine at codon 410 is highly conserved considering 14 species up to frog (Alamut software v2.9), although several species of fish have a serine at this position, suggesting this change is evolutionary tolerated. Likewise, computational analyses suggest this variant does not have a significant effect on KCNJ2 protein structure/function (SIFT: tolerated, PolyPhen2: benign). However, based on the available information, the clinical significance of the p.Asn410Ser variant cannot be determined with certainty. -
