Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ACTN2 protein function. The Asp450Tyr variant in the ACTN2 gene has not been reported as a disease-causing mutation or as a benign polymorphism to our knowledge. Asp450Tyr results in a semi-conservative amino acid substitution of a negatively charged Aspartic acid with a neutral, polar Tyrosine at a position that is conserved across species. In silico analysis predicts Asp450Tyr is damaging to the protein structure/function. The Asp450Tyr variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Nevertheless, no mutations in nearby codons have been reported in association with cardiomyopathy. The variant is found in DCM panel(s). -
