Primary ciliary dyskinesia 7 Pathogenic:4
The DNAH11 c.13069C>T; p.Arg4357Ter variant (rs775720394), also known as c.13090C>T p.Arg4364Ter for NM_003777, is reported in the literature in a compound heterozygous individual affected with primary ciliary dyskinesia and situs inversus (Shoemark 2018, Shoemark 2021). This variant is reported as pathogenic in ClinVar (Variation ID: 454655). This variant is only observed on two alleles in the Genome Aggregation Database (v2.1.1), indicating it is not a common polymorphism. This variant induces an early termination codon and is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay. Based on available information, this variant is considered to be pathogenic. References: Shoemark A et al. Based on the classification scheme VCGS_Germline_v1.3.4, this variant is classified as Pathogenic. Following criteria are met: 0102 - Loss of function is a known mechanism of disease in this gene and is associated with primary ciliary dyskinesia 7, with or without situs inversus (MIM#611884). (I) 0106 - This gene is associated with autosomal recessive disease. (I) 0201 - Variant is predicted to cause nonsense-mediated decay (NMD) and loss of protein (premature termination codon is located at least 54 nucleotides upstream of the final exon-exon junction). (SP) 0251 - This variant is heterozygous. (I) 0304 - Variant is present in gnomAD <0.01 for a recessive condition (v2 & v3: 5 heterozygotes, 0 homozygotes). (SP) 0701 - Other variants predicted to cause NMD comparable to the one identified in this case have very strong previous evidence for pathogenicity (DECIPHER). (SP) 0802 - This variant has moderate previous evidence of pathogenicity in unrelated individuals. It was regarded as pathogenic by multiple diagnostic laboratories in ClinVar. (SP) 1206 - This variant has been shown to be paternally inherited (by segregation analysis). (I) Legend: (SP) - Supporting pathogenic, (I) - Information, (SB) - Supporting benign -

Primary ciliary dyskinesia Pathogenic:1
This sequence change creates a premature translational stop signal (p.Arg4357*) in the DNAH11 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DNAH11 are known to be pathogenic (PMID: 18022865, 20513915, 22184204). This variant is present in population databases (rs775720394, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with DNAH11-related conditions. ClinVar contains an entry for this variant (Variation ID: 454655). For these reasons, this variant has been classified as Pathogenic. -
