This nonsense mutation is categorized as deleterious according to ACMG guidelines (PMID:18414213). It was found in three unrelated probands in our laboratory, twice confirmed de novo. Patients showed intellectual disability, microcephaly, and two also dysmorphic features and seizures -
For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 162158). This premature translational stop signal has been observed in individual(s) with syndromic intellectual disability (PMID: 25920557, 28053047). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Arg437*) in the DYRK1A gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DYRK1A are known to be pathogenic (PMID: 25944381). -
