The FLNC gene encodes Filamin C protein in cardiac and skeletal muscle. Due to study of Rene L Begay et al., 2018 (PMID: 30067491), FLNC variants present in 2.2% of Dilated Cardiomyopathy (DCM) families characterized by a high risk of life-threatening ventricular arrhythmias and a pathological cellular phenotype partially overlapping with arrhythmogenic right ventricular cardiomyopathy. -

The p.G406S variant (also known as c.1216G>A), located in coding exon 8 of the FLNC gene, results from a G to A substitution at nucleotide position 1216. The glycine at codon 406 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
