This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 185 of the ACTA2 protein (p.Arg185Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with ACTA2-related conditions (PMID: 19409525, 25759435; internal data). ClinVar contains an entry for this variant (Variation ID: 381615). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt ACTA2 protein function with a negative predictive value of 80%.
