Loss-of-function variants in COL2A1 are known to be pathogenic (PMID: 20179744). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with autosomal dominant Stickler syndrome and/or high myopia (PMID: 1677770, 12544472, 26747767). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 17355).
