The encoded enzyme is an (alpha)2 dimer which belongs to the class II family of tRNA synthetases. It has been shown to be a target of autoantibodies in the human autoimmune diseases, polymyositis or dermatomyositis. The GARS1 c.236G>A; p.Arg79Gln variant (rs369466037) is reported in the literature in an individual affected with Charcot-Marie-Tooth disease, although its clinical significance was not demonstrated (Volodarsky 2021). This variant is found in the non-Finnish European population with an allele frequency of 0.02% (23/128,638 alleles) in the Genome Aggregation Database (v2.1.1). Computational analyses are uncertain whether this variant is neutral or deleterious (REVEL: 0.69). Due to limited information, the clinical significance of this variant is uncertain at this time.
