GARS1 (HGNC:4162): (glycyl-tRNA synthetase 1) This gene encodes glycyl-tRNA synthetase, one of the aminoacyl-tRNA synthetases that charge tRNAs with their cognate amino acids. The encoded enzyme is an (alpha)2 dimer which belongs to the class II family of tRNA synthetases. It has been shown to be a target of autoantibodies in the human autoimmune diseases, polymyositis or dermatomyositis. The GARS1 c.236G>A; p.Arg79Gln variant (rs369466037) is reported in the literature in an individual affected with Charcot-Marie-Tooth disease, although its clinical significance was not demonstrated (Volodarsky 2021). This variant is found in the non-Finnish European population with an allele frequency of 0.02% (23/128,638 alleles) in the Genome Aggregation Database (v2.1.1). Computational analyses are uncertain whether this variant is neutral or deleterious (REVEL: 0.69). Due to limited information, the clinical significance of this variant is uncertain at this time.
