ClinVar contains an entry for this variant (Variation ID: 132755). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. The NBN c.1373A>G (p.Tyr458Cys) variant has been reported in the published literature in an individual with breast cancer as well as a reportedly healthy individual (PMID: 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared/genes/NBN)). This variant has also been reported in large pan-cancer screening study as a variant of uncertain significance (PMID: 36346689 (2023)). The frequency of this variant in the general population, 0.000004 (1/251184 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -

This variant is reported in 0.00088% of alleles in individuals of European (Non-Finnish) descent in gnomAD and is classified as a variant of uncertain significance in ClinVar (https://www.ncbi.nlm.nih.gov/clinvar/variation/132755/). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -

The p.Y458C variant (also known as c.1373A>G), located in coding exon 10 of the NBN gene, results from an A to G substitution at nucleotide position 1373. The tyrosine at codon 458 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
