The p.G203R variant (also known as c.607G>A), located in coding exon 6 of the SDHB gene, results from a G to A substitution at nucleotide position 607. The glycine at codon 203 is replaced by arginine, an amino acid with dissimilar properties. This variant has been detected in a patient diagnosed with a pheochromocytoma at the age of 60 (Erlic Z et al. Clin Cancer Res. 2009 Oct 15;15(20):6378-85). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
Hereditary pheochromocytoma-paraganglioma Uncertain:1
This missense variant replaces glycine with arginine at codon 203 of the SDHB protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with pheochromocytoma in the literature (PMID: 19825962). This variant has been identified in 2/251304 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
