⢠The p.Ile348Thr variant in the SLC22A5 gene has been previously reported in 1 individual referred for possible carnitine deficiency without a second variant identified (Frigeni et al., 2017). ⢠The p.Ile348Thr variant has also been identified in 10/19,954 East Asian chromosomes by the Genome Aggregation Database (http://gnomad.broadinstitute.org/). Although this variant has been seen in the general population, its frequency is low enough to be consistent with a recessive carrier frequency. ⢠Computational tools predict that this variant does not impact protein function; however, the accuracy of in silico algorithms is limited. ⢠These data were assessed using the ACMG/AMP variant interpretation guidelines. This variant was observed as part of a predisposition screen in an ostensibly healthy population. A literature search was performed for the gene, cDNA change, and amino acid change (where applicable). No publications were found based on this search. Allele frequency data from public databases did not allow this variant to be ruled in or out of causing disease. Therefore, this variant is classified as a variant of unknown significance. -
This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 348 of the SLC22A5 protein (p.Ile348Thr). This variant is present in population databases (rs150544263, gnomAD 0.05%). This missense change has been observed in individual(s) with clinical features of primary carnitine deficiency (PMID: 17486650, 28841266). ClinVar contains an entry for this variant (Variation ID: 25407). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SLC22A5 protein function.
