Inborn genetic diseases Uncertain:1
The c.1970A>G (p.D657G) alteration is located in exon 16 (coding exon 16) of the ADAM17 gene. This alteration results from a A to G substitution at nucleotide position 1970, causing the aspartic acid (D) at amino acid position 657 to be replaced by a glycine (G). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Inflammatory skin and bowel disease, neonatal, 1 Uncertain:1
This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 657 of the ADAM17 protein (p.Asp657Gly). This variant is present in population databases (rs144657795, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with ADAM17-related conditions. ClinVar contains an entry for this variant (Variation ID: 539949). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
