This sequence change does not appear to have been previously described in patients with SDHA-related disorders and has been described in the gnomAD database with a low population frequency of 0.015% (dbSNP rs377470390). The p.Ser52Phe change affects a highly conserved amino acid residue located in a domain of the SDHA protein that is not known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Ser52Phe substitution. Due to these contrasting evidences and the lack of functional studies, the clinical significance of the p.Ser52Phe change remains unknown at this time. -

The p.S52F variant (also known as c.155C>T), located in coding exon 3 of the SDHA gene, results from a C to T substitution at nucleotide position 155. The serine at codon 52 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
