The c.333+5G>C intronic variant results from a G to C substitution 5 nucleotides after coding exon 3 in the BMPR1A gene. This variant was reported in an individual with features consistent with BMPR1A-related juvenile polyposis syndrome (Ambry internal data). This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
Juvenile polyposis syndrome    Uncertain:1 
This sequence change falls in intron 5 of the BMPR1A gene. It does not directly change the encoded amino acid sequence of the BMPR1A protein, but it affects a nucleotide within the consensus splice site of the intron. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a BMPR1A-related disease. Nucleotide substitutions within the consensus splice site are relatively common causes of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of nucleotide changes on RNA splicing suggest that this variant may alter RNA splicing, but this prediction has not been confirmed by published transcriptional studies. In summary, this variant is a novel intronic change with uncertain impact on splicing. It has been classified as a Variant of Uncertain Significance. -
