The POLE c.4522C>G variant is predicted to result in the amino acid substitution p.Arg1508Gly. This variant has been reported in an individual with immunodeficiency (Maffucci et al. 2016. The arginine at codon 1508 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species.
